Studies on the shigella flexneri phage General properties of phage


Abstract
The author has investigated the properties of a phage of which host cell is shigella flexneri la and obtained the following results.
The phage EOP appeared on the various bacilli of shigella flexneri group is resemble to that appeared on the host bacteria and the EOP appeared on the other higella strains except flexneri group is lower grade than that appeared on host cell. And also, in some dysentery bacilli except flexneri group, the phage can not completely multiply. On the spot test in other bacteria except dysentery bacilli, only one strain of E. coli is positive and the other enteric bacilli and cholera vibrio are all negative.
The phage EOP appeared on the various shigella strains except flexneri group is different grade respectively according to each strain. The phage plaques produced in the bacteria of shigella flexneri group are almost same appearance as that produced in the host bacteria and the plaques appeared in other dysentery baiclli except flexneri group, especially in the bacteria of shigella boybii group, are smaller than that produced in host bacteria and, in some cases, the plaques are also not clear.
The adsorption velocity of phage to the various bacilli of shigella flexneri group is almost similar to that to the host bacteria and the phage adsorption, however, to the other shigella strains except flexneri group is very slower than that to host cell. The adsorption of phage to host bacteria is excellent at 37¡ÆC and the adsorption velocity become slower according to decreasing of temperature.
In normal conditions, the latent period, brust size and complete phage liberating time of the phage is 28 minutes, about 50 and 34 minutes respectively. In the bacteria of shigella flexneri group, these characters are almost samely appeared but, in the other shigella strains except flexneri group, the latent period, burst size and complete phage liberating time become considerably retarded or lowered than that in host bacteria.
The phage change to host range mutant according to passing through the bacteria of which antigenic properties are different from the host cell. The velocity constant of phage neutralization of the antiserum is about 128 min.-1 in normal condition and about 50% of phage is inactivated by keeping of the phage at 50¡ÆC for 80 minutes. The appearances of phage inactivation by anti-phage serum and heat are evenly the first order kinetics.
The phage is relatively stable at pH 6.0~9.0 and about 60% of phage is inactivated
by the sonic vibration for 2 minutes. And the phages inactivated by ultraviolet ray produce the photoreactivation but not the multiplicity reactivation.
